The advantages to reading an assay in kinetic mode rather than with a single endpoint read are numerous. It makes it possible to work within the initial reaction rate with very low substrate turnover. It enables the use of assays with very low signal and short linearity of reaction progress curves for hit identification. Also, it dramatically reduces physical interference from compounds in the screening collection. Historically, it has been very difficult to run these assays in HTS labs due to long read times and issues of data management for multiple reads. We have leveraged technology to make kinetic read for HTS a routine process. We will present how this has been achieved at BMS and give examples which clearly demonstrate the advantages of kinetic-mode screening.
GTCbio's 2nd Assay Development & High Throughput Screening Conference on May 31-June 1 will take place at the Omni Parker House in downtown Boston, MA. The meeting will be held concurrently with GTCbio's 2nd Protein Kinases in Drug Discovery Conference.
